Nous cherchons constamment à améliorer les protocoles de production des vecteurs (approches moléculaires, cellulaires et virologiques) dans le but :

  • d’augmenter la qualité des vecteurs produits,
  • de réduire les coûts et les délais de production,
  • et d’étendre nos services.

Les adénovirus sont des agents pathogènes courants dans la population humaine. Ils persistent également pendant des décennies. Par conséquent, l'injection de vecteurs Ad chez l'homme peut être compliquée. Nos objectifs de R&D sont de comprendre comment ces vecteurs sont ciblés par des protéines extracellulaires qui peuvent modifier leur immunogénicité dans les tissus humains.  Notre objectif est ainsi d'essayer de prédire les effets indésirables qui pourraient survenir lorsque les vecteurs sont injectés dans le cerveau. Cela permettra d’élargir notre place sur la scène internationale en donnant accès à des vecteurs CAV-2 robustes pour le transfert de gène non seulement chez l’animal, mais également chez l’homme. Les projets de R&D font l'objet de collaborations internationales entre le plateau IGMM de la plateforme et des laboratoires implantés aux Pays-Bas, au Royaume-Uni et aux États-Unis.

Dans le cadre des programmes de R&D Biocampus Montpellier, le plateau IGF de la plateforme développe un projet conjoint avec l’équipe de Carole Crozet (Institut des Neurosciences de Montpellier).

Le but de ce projet est de mettre en place une approche permettant d’augmenter l’efficacité d’édition du génome de cellules souches pluripotentes induites humaines (hiPSCs) comparativement aux méthodes utilisées habituellement, tout en diminuant les effets non-spécifiques de ces dernières. L’approche que nous développons actuellement se base sur l’utilisation de nanoparticules dérivées des rétrovirus murins (Mangeot et coll., 2019). Ce type de vectorisation permet de délivrer directement le complexe Cas9/ARN guide (gRNA) dans les cellules. Cette approche sera couplée avec l’utilisation d’un AAV (Adeno-associated virus) pour introduire l’ADN donneur afin d’induire la modification génétique désirée par recombinaison homologue (Martin and coll. 2019 and Chen and coll., 2019).
