Lentivirus and Retrovirus Production and Purification

The lentivector production system used at the PVM platform is based on the 2nd generation HIV-1 derivative, with particles pseudotyped with the VSV-G envelope glycoprotein. The production of murine retroviruses is also available upon request.

Basic Service Offered

• Viral particle production by triple transfection in 293T cells
• Purification and concentration (100×) by ultracentrifugation on a sucrose gradient
• Physical titration: ELISA p24
• Infectious titration if the vector contains a fluorescent reporter gene under a ubiquitous promoter (analysis by flow cytometry after transduction)

Available Production Formats

• Large scale: 1 virus → 1400 µL
• Small scale: 2 different viruses → 700 µL/virus

Optional Services on Request

• Production of non-concentrated viral supernatants
• Alternative concentration methods: PEG precipitation or filtration
• Production of vectors derived from murine retroviruses MuLV pseudotyped with VSV-G
